Detection

Biocare Medical Offer a large range of detection systems for the immunohistochemistry scientist. See which ones suit your needs:

4 plus

A sensitive two-step HRP and AP detection method

Biocare's 4plus detection systems are designed for reliable, cost-effective, two-step detection, providing a high-level of sensitivity. The vast majority of primary antibodies can be diluted two-fold compared to other commercially available avidin-biotin detection systems. Biocare's prediluted antibodies are all standardized to work with the 4plus detection system.

4plus detection system can be used with mouse and rabbit primary antibodies. After labeling the antigen with primary antibody, a species-specific or universal, affinity-purified, biotinylated secondary antibody is added to bind. Horseradish peroxidase (HRP) or alkaline phosphatase (AP) labeled-streptavidin is then added. A chromogen/substrate is applied that reacts with a HRP or AP to produce an intense color signal.

Chromogens

Biocare Medical offers the largest variety of IHC-specific permanent chromogens, for use with the two most commonly used enzyme labels: Horseradish Peroxidase (HRP) and Alkaline Phosphatase (AP). Chromogens are chemical compounds used within IHC staining procedures that produce a colored end-product that can be visualized under bright-field microscopy.

Fluorescence

Biocare's Signal Amplification Reagent increases fluorescent signals 3 to 5-fold when used in combination with DyLight immunofluorescent detection kits. This increased sensitivity is perfect for identification of low abundance proteins as well as routine work. Amplify your staining intensity for nuclear, cell membrane, and cytoplasmic antigens with Biocare's Fluorescence Enhancement Probe technology. For use on human tissue only.

  • 3 to 5-fold Signal amplification with Biocare DyLight Detection Kits
  • Superior to FITC and TRITC
  • Multiplex detection protocols

MACH 1™

The MACH 1 Universal Polymer Detection system offers a cost-effective alternative method of polymer detection for IHC laboratories (not available in the U.S.). Specifically titered for Biocare prediluted antibodies, MACH 1 provides superior staining and a reduced protocol time as compared to the conventional ABC system.

This new two-step, biotin-free technology uses a specific probe to detect mouse antibodies. The probe is followed by an HRP-polymer that binds to both the probe and to rabbit antibodies. Time consuming avidin-biotin and normal serum blocking steps are eliminated. The innovative HRP-polymer technology provides superior staining for mouse and rabbit antibodies when compared to ABC, biotin-streptavidin (BSA) and other polymer detection kits.

MACH 2™

MACH 2 Detection is a one-step / one-solution, biotin-free detection system which combines superior work flow and sensitivity for mouse and rabbit primary antibodies.

MACH 2 detection consists of a single reagent applied after the primary antibody. MACH 2 is ideal for use with prediluted antibodies or concentrates with equal success. MACH 2 may be used for 7-step multiplex stains with certain antibodies.

MACH 2 increases staining specificity compared to conventional dextran backbone polymer. MACH 2 is available in anti-mouse, anti-rabbit, and 'universal' (anti-mouse & anti-rabbit) formulations, labeled with either Horseradish Peroxidase (HRP) or Alkaline Phosphatase (AP).

  • Increase specificity 3-4 fold in nuclear staining
  • Increase specificity 2 fold in cytoplasmic/cell surface staining
  • Superior to dextran backbone polymers

MACH 3™

MACH 3 is a two-step, biotin-free detection system which provides excellent specificity, sensitivity and nuclear staining for mouse or rabbit primary antibodies.

MACH 3 Features:

  • A sensitivity-enhancing secondary reagent
  • Increased density of enzymes bound to tertiary target
  • 5- to 10- fold increased staining sensitivity when compared to conventional dextran polymer detection kits
  • Superior staining for nuclear and cytoplasmic/cell surface antigens
  • Elimination of background
  • High primary antibody dilution significantly improves specificity and reduces cost
  • Adapted for all Biocare's prediluted antibodies
  • Compatible with automated immunostainers

MACH 4™

MACH 4 is a highly sensitive universal detection system. The MACH 4 system can detect either mouse or rabbit antibodies, and is a two-step universal detection method.

MACH 4 micro-polymer system incorporates two key technologies:

Enhanced probe to increase sensitivity 10- to 20- foldIncreased density of enzymes bound to tertiaries

MACH 4 Features:

  • 10 - 20 times more sensitive than conventional dextran polymer systems
  • 20 - 40 times more sensitive for nuclear staining
  • Suitable to detect mouse specific anti-biotin or antidigoxigenin antibody in in situ hybridization (ISH)
  • Micro-polymer allows superior specificity/sensitivity, nuclear access and primary antibody dilution
  • Compatible with automated immunostainers

Multiplex

Biocare Medical is the proven leader in providing multiplex detection platforms that enable simultaneous IHC staining with multiple antibodies on a single slide. This superior detection system increases reimbursement per slide, simplifies procedures and reduces turnaround time.

Multiplex Micro-polymer Detection Kits

The innovative AP (Alkaline Phosphatase) and HRP (Horseradish Peroxidase) micro-polymer technology is specially designed for a rapid multiplex stain procedure, i.e. multiple antigens on the same slide. The micro-polymer confers significant increase in staining sensitivity when compared to conventional polymer detection systems. The micro-polymer multiplex detection kits are optimized for human tissue.

Key features of multiplex micro-polymer detection:

  • Simplifies protocols by reducing the number of steps (4 steps)

  • Eliminates endogenous biotin blocking

  • Process >50 percent fewer slides

  • Results in less than two hours

  • Compatible with automated IHC instruments

  • Significant increase in staining sensitivity

PromARK™

Biocare's PromARK micro-polymer detection systems are designed specifically for use on animal tissues. The micro-polymer technology provides superior sensitivity and minimum cross-reactivity to endogenous mouse and rat IgG. The Promark product line includes specially formulated heat-retrieval solutions, blocking agents, and one- and two-step detection reagents.

  • High staining sensitivity and specificity
  • Minimum cross-reactivity to endogenous IgG with proprietary blocking reagents
  • Elimination of endogenous biotin
  • Reduced number of IHC steps
  • Use with formalin-fixed, paraffin-embedded tissues, floating sections, frozen sections and cell culture preparations
  • Highly effective ancillary reagents
  • Suitable for both manual and automated techniques

Animal tissues contain endogenous immunoglobulins that produce significantly high levels of background staining when standard anti-mouse/anti-rabbit detection systems are employed. Biocare provides micro-polymer detection systems for use with primary antibodies from various sources (mouse, rat, rabbit, goat) on a range of tissues (mouse, rat, canine, feline, bovine, equine, porcine, ovine) which result in reduced background staining and higher sensitivity.

Starr Trek™

Available for use with HRP or AP, these highly cost-effective kits are compatible with Biocare prediluted antibodies. Using a streptavidin AP or HRP reduces non-specific background staining. Starr Trek is much more stable than the ABC complex.

Kits are color-coded for ease of use. Starr Trek kits come with Universal Link and avidin-enzyme reagents. We recommend Betazoid DAB for optimal performance with the HRP Kit.
