ELISAs (Enzyme-linked immunosorbent assays) are routinely used for the qualitative and quantitative assessment of cytokines, chemokines, growth factors, phosphorylated targets, immunoglobulins and other immunological markers. PrimeFlow™ RNA Assay - Enter a new dimension of single-cell analysis
PrimeFlow™ RNA Assay reveals the dynamics of RNA and protein expression within individual cells, facilitating unprecedented analysis of their correlation as the cell changes over time or in response to stimulus. This novel assay employs a proprietary fluorescent in situ hybridization (FISH) technique enabling simultaneous detection of up to three RNA transcripts in a single cell using a standard flow cytometer. RNA detection may be teamed together with intracellular and cell surface antibody staining to elevate the understanding of single cell dynamics to a new dimension.
Development of the assay is based upon proven and well-published Affymetrix® ViewRNA™ FISH assays designed for microscopic analysis of RNA in cells and tissues. The assay combines paired oligonucleotide probe design with branched DNA (bDNA) signal amplification to robustly detect gene expression at the single-cell level.
Coupling RNA expression with protein detection on a flow cytometer generates multiparametric data in heterogeneous cell populations and offers in-depth and high content details at the single cell level. In contrast, microarrays and sequencing can provide comprehensive gene expression data in bulk sample preparations; however, such bulk sample preps can mask the individual effects of different cellular subsets. Using the PrimeFlow™ RNA Assay, specific cell populations may be analyzed for unique transcript expression levels or cell subsets evaluated over time to determine transcriptional regulation and protein expression simultaneously. Such unique and valuable insights are highly applicable to answering previously unanswerable questions and have broad implications for advancing research across multiple fields of biology.

QuantiGene is a signal amplification technology utilized for the quantitation of gene expression transcripts. We have 4 different technologies that incorporate the branched DNA technology which is used to amplify the signal and provides a more robust and accurate measurement compared to qPCR. The QuantiGene Plex and QuantiGene Singlelex assays are designed to quantitatively measure RNA directly from cell or tissue homogenates without RNA purification. PrimeFlow™ and ViewRNA are in situ based assays that can measure or visualize RNA directly in the cells.

Immunohistochemistry and Immunocytochemistry
Microscopy is the most widely used imaging method to explore biological systems, in which immunocytochemistry (ICC), the localization of proteins within cells, and immunohistochemistry (IHC), which reveals the abundance, distribution, and localization of biomarkers within a tissue, can be viewed directly. These techniques provide insight into cellular structure and mechanisms and are applicable for basic research, in addition to being indispensable in clinical settings. Target antigens may be evaluated using specific antibodies directly conjugated with an enzyme or fluorophores, or indirectly using similarly labeled secondary antibodies and reagents.
Immunoprecipitation and Western Blot
Immunoprecipitation (IP) is an enrichment procedure of specific proteins, protein modifications, or protein motifs from a heterogeneous protein sample such as cell lysate. The enrichment occurs through the use of an antibody that commonly recognizes a protein in its native or semi-native format. IP is often followed by a western blot for analysis, but can be followed by other applications as well. IP allows for the detection of proteins of low abundance, protein:protein interactions and protein post-translational modifications (PTMs).
Western Blot (WB)
Western Blot (WB), also referred to as immunoblot, is a common, effective method for determining relative protein or protein-modification levels in a sample. Denatured proteins are separated via SDS-PAGE and transferred to a membrane where the proteins are detected via the use of antibodies. The relative intensity, and location of the protein on the blot, allow for its identification and quantification.
