ISOFLUX OVERVIEW

The IsoFlux System enables molecular analysis of circulating tumor cells (CTCs) and other rare cell types. Using Fluxion's proven microfluidic technology, the platform recovers CTCs from biological samples with the highest level of sample integrity. The recovered cells move immediately to downstream molecular analysis including mutation detection, next-generation sequencing, and gene expression.

Backed by a proven microfluidic technology platform.

The IsoFlux System uses proprietary CellSpot Technology to deliver enriched cells in a format ideal for next generation molecular applications. CellSpot increases the purity, integrity, and yield of isolated cells, while also making them easy to access for use in downstream applications.


IsoFlux CTC Sample Prep1. SAMPLE PREPARATION

A tube (7.5-10mL) of peripheral blood is collected and sent to processing site. The blood is centrifuged against a density gradient and PBMC layer is retrieved. Immunomagnetic particles are added to the sample that target the cells of interest.

Not just limited to EpCAM…IsoFlux samples can be processed with one or multiple isolation markers defined by the user.

The sample is then transferred into a microfluidic cartridge. The sample flows from the inlet well across an isolation zone, where the target cells are sequestered. The remaining non-target cells flow into a waste reservoir.

Within the isolation zone, the sample flows up into a cavity. A permanent magnet is placed on the roof of the channel to attract the labeled target cells.

The uniformity of the laminar flow profile and magnetic field gradient ensures high cell recovery & minimal background contamination.

Once the target cells are captured and washed, the disc that forms the roof of the channel is removed from the cartridge inside the instrument. The target cells are aspirated and transferred into a microfuge tube for further analysis. This approach is optimal for molecular analysis because the cell recovery off of the chip is high (>90%) and the dilution volume is very low (<20uL).

The enriched sample is immediately ready for downstream analysis usingmicroscopy, PCR, qPCR, FISH, NGS, and microarrays.
