Count every responding cell

ELISpot is best for counting individual protein-secreting cells. Live immune cells are added to a PVDF plate coated with a capture antibody. When stimulated, the cells release their target protein, which is captured close to each cell. After the cells are removed, a detection antibody and enzyme are added, and a precipitating substrate forms a visible spot at each secretion site, with each spot representing a single responding cell.
