MHC Tetramers and Monomers

MBL offers an expansive product portfolio for antigen specific T-cell detection.  Products include MHC tetramers, monomers, peptides, and their innovative QuickSwitch™ kit for creating your own custom tetramers or screening peptide-MHC affinities. New alleles are continuously added.  They offer MHC tetramer products for species including Human, Mouse, Cyno Macaque, Rhesus Macaque, Chimera, and Chicken.

MHC Tetramers

The ability to quantitate a T-cell response is an important piece of information for better understanding of various questions such as efficacy of new vaccines/therapies, evaluating disease progression or clearance, and many other questions.  T cell antigen receptors (TCR) recognise peptides that are expressed on antigen presenting cells.  Antigen specific T cell responses can be analysed by using MHC tetramer molecules, where each monomer is bound with a specific peptide.  This method provides more robust T cell information compared to only using antibodies because the MHC tetramers will only bind to T cells that recognize the peptide sequence.  Using this method, it is certain that any signal that is observed is due to a specific T cell response and not due to an overall increase in T cell presence.

MHC tetramers are composed of four monomers, each with a specific peptide.  The monomers are biotinylated and held together using  fluorescently-labelled streptavidin.  Some researchers have found using MHC multimers with more than 4 monomers result in non-specific binding.  By using MHC tetramers, binding affinity of the peptide and TCR remains high, while reducing/avoiding low non-specific binding.

Biotinylated MHC monomers, the building blocks of MHC tetramers (Class I or Class II), are now available for tetramer construction flexibility and novel applications.

Peptides

MBL’s stock and custom peptides can be used for stimulation assays or with custom tetramer generation using the QuickSwitch™ custom tetramer kits. All peptides undergo a rigorous quality control process to ensure they are high grade including MS, LC-MS and HPLC analysis as well as COA documentation to ensure at least 95% purity.

Their stock peptides are specific to the MHC tetramer product line and contain peptides for popular disease targets such as CMV, MuLV, melanoma, prostate cancer and other virology and cancer targets.

Peptide lengths can range from 2 amino acids to 100 amino acids depending on the peptide of interest, and are available in sizes from 1mg- 500mg and above.
