Target enrichment through a solution-based capture system enables enrichment of defined regions of interest up to several Mb in size. Roche NimbleGen's SeqCap EZ enrichment technology, combined with PacBio's SMRT Sequencing, has demonstrated the ability to use 6 kb fragments and is applicable to practically any type of SeqCap EZ panel. They have demonstrated efficient enrichment for the Major Histocompatiblity Complex and a comprehensive cancer gene panel. On average, 61% of the reads were on target and an average enrichment factor of 1300x was obtained. With PacBio long reads, heterozygous SNPs were used to phase the reads and generate haplotypes.

Direct PCR amplification of a specific region is a powerful technique for performing targeted sequencing. When combined with multiplexing capabilities, PCR amplification is an efficient approach to studying many samples. PacBio offers two barcoding strategies for multiplexing. In the first approach, barcodes are added to the end of the amplicon during PCR, and the standard SMRTbell adapter is subsequently ligated onto the barcoded amplicon.
