beata.jpg Widefield Microscopy as a Screening Tool for Three- Dimensional Cell Culture

Within the body, cells are typically embedded inside ECM or are in direct physical contact with cells from either the same or different lineages.  And today there is an ever growing body of evidence that suggests that Three-dimensional (3D) cell culture systems, represent more accurately the actual microenvironment where cells reside in tissues, compared with 2D culture systems.  Therefore the behaviour 3D-cultured cells is more reflective of in vivo cellular responses.  This has motivated the development of in vitro cell culture platforms that better represent the cellular microenvironment.

Different biological readouts from 3D cell models have been described including the analysis of cell lysates using analytical methods, such as plate reader-based luminescence or fluorescence measurements. However, high throughput widefield imaging methods have been demonstrated as promising techniques for morphological characterization and phenotypic screening in various 3D culture models.

BioTek  have released a new application guide introducing various imaging assays performed with  BioTek's  high-content imaging instruments and image analysis software to characterize phenotypic changes in cancer spheroids in response to different compound treatments, including:
